FLAG peptide is a peptide composed of eight amino acids (sequences: Asp-Tyr-Lys-Asp-Asp-Asp-Asp-Lys), which has an intestinal kinase cleavage site. FLAG peptide is used for antibody mediated recognition and purification of recombinant proteins.
FLAG in Vitro Experiment:
Fusion protein technology has become an important tool for solving numerous problems related to recombinant protein production. The characteristics of additional labeling facilitate identification and provide a one-step purification process for fusion proteins through appropriate matrix columns using cell extracts or supernatants. Labeled peptides allow for elution under non denaturing conditions.
This program uses antibodies M2 and M5. Firstly, describe the marker as a calcium dependent epitope of a monoclonal antibody. It is a highly acidic octapeptide that can fuse with proteins of interest at the n-terminus. As a highly hydrophilic peptide segment, labeling has a high surface probability. In the presence of Ca2+, the immunoaffinity column can capture the Flag fusion protein and elute it through EDTA at low concentrations, neutral pH, and near physiological conditions.
