In vitro study:
Sodium dichloroacetate can induce apoptosis of human lung, breast and brain cancer cells. After Sodium dichloroacetate treatment, ROS levels in cancer cells increased, MMP depolarization, and apoptosis increased in vivo and in vitro. Sodium dichloroacetate inhibits the activity of pyruvate dehydrogenase kinase (PDK) and thus stimulates mitochondrial pyruvate dehydrogenase (PDH). When PDH is off, PDH no longer converts pyruvate to Chemicalbook acetyl-CoA (necessary for mitochondrial respiration and glucose-dependent oxidative phosphorylation processes). Thus, Sodium dichloroacetate converts cellular metabolism from glycolysis to glucose oxidation, reduces mitochondrial membrane potential, and promotes the opening of mitochondrial transport pores. This series of metabolic switches contributes to the translocation of pro-apoptotic mediators such as cytc and AIF. Therefore, Sodium dichloroacetate can drive cancer cells to commit suicide through apoptotic reaction.

In vivo study:
Sodium dichloroacetate acts as a cell growth inhibitor in vivo and in vitro without causing programmed cell death (apoptosis). Sodium dichloroacetate is a highly safe drug with no heart, lung, kidney or bone marrow toxicity. The most serious side effects include peripheral neuropathy, which is a reversible process. Sodium dichloroacetatehas anticancer activity in several cancer types, including colon, prostate, ovarian, neuroblastoma, lung, cervical, endometrial, cholangiocarcinoma, sarcoma, and T-cell lymphoma. Other antitumor effects of Sodium dichloroacetate have also been reported, including obstruction of angiogenesis, alteration of HIF-1α expression, alteration of pH regulators V-ATPase and MCT1, and alteration of other cell survival regulators such as PUMA, GLUT1, Bcl2, and p53. In a rat model of highly metastatic breast cancer, Sodium dichloroacetate can significantly reduce lung metastatic burden. In C57BL/6 mice, in vivo administration of Sodium dichloroacetate-Na induced a 20% improvement in survival and reduced tumor diameter, volume, and weight without affecting their weight and avoiding metastasis.
