Brief introduction

Soluble in ethanol and water, slightly soluble in ethyl acetate and benzene, insoluble in ether and carbon tetrachloride, corrosive to copper and aluminum, and irritating chemicals
Application
Tris is widely used in the preparation of buffer solutions in biochemical and molecular biology experiments. For example, the commonly used TAE and TBE buffer solutions (used for nucleic acid dissolution) in biochemical experiments require Tris; Because it contains amino group, it can undergo Condensation reaction with aldehyde. The pH of Tris alkali aqueous solution is around 10.5. Generally, hydrochloric acid is added to adjust the pH value to the desired value to obtain a buffer solution with that pH value. However, attention should also be paid to the effect of temperature on the pKa of Tris. Since Tris buffer is a weakly alkaline Chemicalbook solution, DNA will be Deprotonation in such a solution, thus improving its solubility. People often add EDTA to Tris hydrochloric acid buffer to make "TE buffer", and TE buffer is used to stabilize and store DNA. If the acid solution used to adjust the pH value is replaced with acetic acid, a "TAE buffer" (Tris/Acetate/EDTA) is obtained, while replacing it with boric acid results in a "TBE buffer" (Tris/Borate/EDTA). These two types of buffers are commonly used in nucleic acid electrophoresis experiments.



Preparation of buffer solution
A Tris HCl buffer solution with a concentration of 6.057g/L and pH=8.8 was prepared by adjusting the pH value of Chemicalbook using trihydroxymethylaminomethane hydrochloride as the solute and deionized water as the solvent.
Advantage
A preparation method of Tris, the specific preparation steps are as follows: (1) adding trimethylolmethane into methanol aqueous solution, heating to 50-70 ºC, stirring and dissolving, wherein the mass volume ratio of the trimethylolmethane to methanol aqueous solution is 8:3-7 in g/ml, and the methanol aqueous solution is prepared by mixing pure water and methanol in a volume ratio of 2:3; (2) Add charcoal activated Chemicalbook carbon to the solution, wherein the weight ratio of charcoal activated carbon to trimethylolmethane is 0.5-2:100, keep it at 45-55 ºC for 20-40 minutes, and then filter while hot to collect the filtrate; (3) Concentrate the filtrate under reduced pressure at a concentration temperature of 70-80 ºC until crystallization occurs, and let it cool down; (4) After the crystallization is separated by suction filtration, it is washed with anhydrous ethanol and dried at 40-60 ºC for 3-5 hours to obtain.
